Development of Disease-Specific Human Organoids and a Drug Discovery Platform for Regenerative Nanomedicine

The human central nervous system possesses only a limited capacity for regeneration, making functional recovery after injury or disease extremely challenging. Understanding the fundamental principles that govern central nervous system regeneration and translating this knowledge into effective therapies therefore remains one of the major challenges in life science. Our laboratory integrates interdisciplinary research and we aim to establish innovative scientific platforms that advance regenerative medicine by harnessing the intrinsic regenerative capacity of human organs to restore function after injury or disease. To achieve this goal, our research is organized around three complementary approaches: 1. Engineering Human Organs Using human induced pluripotent stem cell (hiPSC)-derived organoids—three-dimensional models that recapitulate key features of human organs—we investigate the cellular and molecular principles underlying central nervous system (CNS) development. 2. Modeling Human Disease We combine animal models with human organoids to establish disease models that recapitulate pathological processes. Using these platforms, we investigate the molecular mechanisms underlying CNS disorders, including spinal cord injury, with the aim of identifying key regulators of disease progression and regeneration. 3. Regenerative Nanomedicine and Drug Discovery By integrating nanoscale biomaterials with organoid-based drug screening technologies, we develop novel therapeutic strategies that promote neural tissue regeneration and functional repair. In addition, we establish human organoid-based platforms for evaluating regenerative transplantation therapies, bridging fundamental discoveries with future clinical applications.
